目的:分别建立高效液相色谱-电雾式(CAD)和蒸发光散射(ELSD)2种通用型检测器测定蒺藜皂苷提取物中蒺藜皂苷K含量的方法。方法:采用高效液相色谱法,Waters Symmetry Shield RP18为色谱柱,以乙腈-水为流动相,梯度洗脱,体积流速为1.0 mL·min-1,柱温为25 ℃;使用CAD和ELSD检测器测定,并对样品实测结果进行比较。结果:2种方法线性关系均良好,精密度、重复性、稳定性及加样回收率皆符合方法学要求,CAD的检出限和定量限均比ELSD低。结论:2种检测方法都可用于蒺藜皂苷提取物中蒺藜皂苷K的含量测定,CAD检测器比ELSD检测器具有更高的灵敏度和精密度。 |

Objective: To establish two methods for the determination of Tribulusteristussaponin K in tribulus teristus saponin extract with CAD and ELSD detectors. Methods: HPLC was performed on Waters Symmetry Shield RP18 column with mobile phase composed of acetonitrile-water at a fl ow rate of 1.0 mL·min-1 in gradient elution mode, the column temperature was 25℃. CAD and ELSD detectors were respectively used to detect, and the content determination results of the samples were compared. Results: The linear relationship between the two methods was good, and the precision, repeatability, stability and recovery rate met the methodological requirements. The detection limit and quantitation limit of CAD were lower than those of ELSD. Conclusion: Both methods can be used to determine the content of Tribulusterrestrissaponin K in tribulus terrestris saponin extract. CAD method has higher sensitivity and precision than ELSD method. |
