当归六黄汤出自金代医家李杲的《兰室秘藏》煮散剂,被誉为“治盗汗之圣药”具有滋阴降火,固表止汗之功效。临床疗效明显,但其质量标准物尚不明确,不利于其疗效的提升和质量的保证以及临床的扩展和深化。因此该研究利用文献挖掘、网络药理学多种技术,明确当归六黄汤的质量标志物及其作用机制,从而有效控制质量,确保临床疗效。首先,研究基于中药质量标志物( Q-marker) “五原则”对当归六黄汤质量标志物进行预测分析,提示可选择阿魏酸、梓醇、地黄苷D、黄芩苷、汉黄芩苷、汉黄芩素、小檗碱、黄柏碱、黄芪甲苷作为当归六黄汤的Q-marker。其次,利用中药系统药理学数据库( TCMSP) 、ChEMBL 与SwissTargetPrediction 平台获取功效标志物的潜在作用靶点,构建当归六黄汤功效标志物的蛋白互作网络。利用拓扑、GO、KEGG富集分析构建“关键靶点-信号通路-生物过程”网络,进而阐释当归六黄汤功效标志物的作用机制。网络药理学研究结果表明当归六黄汤功效标志物可能通过作用于MAPK、雌激素、PI3K-Akt等通路从而调节MAPK级联、一氧化氮生物合成过程、细胞凋亡过程。该研究利用文献挖掘结合( Q-marker) “五原则”预测大柴胡汤的功效标志物,并运用网络药理学从分子水平探讨其作用机制,旨在为当归六黄汤质量控制研究提供参考,并为其临床应用提供科学依据。 |

Angelica Liuhuang Decoction is derived from the decoction of Lanshi Secret by Li Gao, a physician in the Jin Dynasty. It is known as "the holy medicine for night sweats", which has the effect of nourishing Yin and reducing fire, strengthening surface and antiperspirant. The clinical effect is obvious, but its quality standard is not clear, which is not conducive to the improvement of efficacy and quality assurance as well as clinical expansion and deepening. Therefore, this study uses multiple techniques of literature mining and network pharmacology to clarify the quality markers of Danggui Liuhuang Decoction and its mechanism of action, so as to effectively control the quality and ensure clinical efficacy. Firstly, based on the "five principles" of traditional Chinese medicine quality marker (Q-marker), the prediction analysis of quality marker of Danggui Liuhuang Decoction was carried out, suggesting that ferulic acid, catalpa, rehmannioside D, baicalin, han baicalin, han baicalin, berberine, phellodendron and astragaloside IV could be selected as Q-marker of Danggui Liuhuang decoction. Secondly, the potential targets of efficacy markers were obtained by using TCMSP, ChEMBL and SwissTarpetPrediction platform, and the eggs of efficacy markers of Danggui Liuhuang Decoction were constructedWhite interaction network. Topological, GO and KEGG enrichment analyses were used to construct a network of "key target-signal pathway-biological process", and then to elucidate the mechanism of efficacy markers of Danggui Liuhuang Decoction. The results of network pharmacological studies indicate that the efficacy markers of Dangguiliuhuang Decoction may regulate the MAPK cascade, nitric oxide biosynthesis and cell apoptosis by acting on MAPK, estrogen and PI3K-Akt pathways. In this study, "Five principles" of Q-marker were used to predict the efficacy markers of Radix buaihu decoction, and network pharmacology was used to explore its mechanism of action from the molecular level, aiming to provide reference for the quality control research of Radix Angelicae Liuhuang Decoction, and provide scientific basis for its clinical application. |
