      目的:利用AlphaLISA方法建立抗白介素-17受体单抗的生物学活性测定方法。方法:以人包皮成纤维细胞系作为靶细胞,通过AlphaLISA检测系统建立抗IL-17R单抗的生物学活性测定方法,根据四参数拟合分析计算抗IL-17R单抗的相对效价,并对该方法进行了验证。结果:抗IL-17R单抗在该方法中存在量效关系,并且符合四参数方程:y=(A-D)/[1+(x/CB]+D。6批抗IL-17R单抗经3次测定,相对效价平均值在(84.93±3.12)%~(107.51±1.66)%,相对标准偏差小于5%。2批回收率样品经3次测定,回收率分别为(100.66±13.65)%和(115.69±3.85)%。结论:本研究利用AlphaLISA方法在国内首次成功建立重复性好、准确性高的抗IL-17R单抗的生物学活性测定方法。
英文摘要:
      Objective: To establish an AlphaLISA method for determining the biological activity of anti-IL-17R monoclonal antibody (mAb). Methods: Human foreskin fibroblast-1 (HFF-1) cell line was used as target cell and the method for determining the biological activity of anti-IL-17R mAb was established by AlphaLISA system. The relative potency of the anti-IL-17R mAb was calculated according to four-parameter fitting analysis. The established method was validated. Results: The anti-IL-17R mAb showed a dose-response relationship and the determination result complied with the following four-parameter equation:y=(A-D)/[1+(x/C)B]+D. Six batches of anti-IL-17R mAb were tested for 3 times. The mean relative potencies were from (84.93±3.12)% to (107.51±1.66)%, with relative standard deviation less than 5%. Two batches of recovery samples were tested for 3 times and the recovery rates were (100.66±13.65)% and (115.69±3.85)% respectively. Conclusion: A novel AlphaLISA method for determining the biological activity of anti-IL-17R mAb was successfully established, which showed good reproducibility and accuracy.
